class=\"wp-block-paragraph\">\u5728\u4f20\u7edf\u65b9\u6cd5\u4e2d\uff0c\u4f7f\u7528\u4e0d\u542b\u4e19\u916e\u9178\/\u5c3f\u82f7\uff08PU\uff09\u7684\u57f9\u517b\u57fa\u8fdb\u884c\u9009\u62e9\u6027\u57f9\u517b\u5bf9\u4e8e\u751f\u6210\u8f6c\u7ebf\u7c92\u4f53\u80de\u8d28\u6742\u79cd\u81f3\u5173\u91cd\u8981\u3002\u5728\u6b64\uff0c\u6211\u4eec\u4ecb\u7ecd\u4e00\u79cd\u5229\u7528\u5fae\u6d41\u63a7\u88c5\u7f6e\u751f\u6210\u8f6c\u7ebf\u7c92\u4f53\u80de\u8d28\u6742\u79cd\u7684\u65b9\u6848\uff0c\u8be5\u65b9\u6848\u5373\u4f7f\u5728\u6dfb\u52a0PU\u7684\u6761\u4ef6\u4e0b\u4e5f\u80fd\u53d1\u6325\u4f5c\u7528\u3002\u6211\u4eec\u63cf\u8ff0\u4e86\u5fae\u6d41\u63a7\u88c5\u7f6e\u7684\u5236\u5907\u3001\u90e8\u5206\u7ec6\u80de\u878d\u5408\uff08\u7ebf\u7c92\u4f53\u8f6c\u79fb\uff09\u4ee5\u53ca\u8f6c\u7ebf\u7c92\u4f53\u80de\u8d28\u6742\u79cd\u7684\u6536\u96c6\u6b65\u9aa4\u3002\u968f\u540e\uff0c\u6211\u4eec\u8be6\u7ec6\u8bf4\u660e\u4e86\u901a\u8fc7\u9650\u5236\u6027\u7247\u6bb5\u957f\u5ea6\u591a\u6001\u6027\uff08RFLP\uff09\u5206\u6790\u786e\u8ba4\u80de\u8d28\u6742\u79cd\u4e2d\u7ebf\u7c92\u4f53DNA\uff08mtDNA\uff09\u91cd\u65b0\u805a\u96c6\u548c\u8fdb\u884cmtDNA\u5206\u578b\u7684\u6d41\u7a0b\u3002<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">1.&nbsp;&nbsp;&nbsp; \u7ebf\u7c92\u4f53\u6742\u5408\u7ec6\u80de\u5236\u5907\u7684\u6280\u672f\u7a81\u7834\uff1a\u4ece\u4f20\u7edf\u65b9\u6cd5\u5230\u5fae\u6d41\u63a7\u9769\u65b0<\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">\u7ebf\u7c92\u4f53\u6742\u5408\u7ec6\u80de\u4f5c\u4e3a\u7814\u7a76\u7ebf\u7c92\u4f53\u529f\u80fd\u4e0e\u75be\u75c5\u5173\u8054\u7684\u91cd\u8981\u6a21\u578b\uff0c\u5176\u4f20\u7edf\u5236\u5907\u9700\u901a\u8fc7\u53bb\u6838\u4f9b\u4f53\u7ec6\u80de\u4e0e mtDNA \u7f3a\u9677\u53d7\u4f53\u7ec6\u80de\u878d\u5408\uff0c\u4f9d\u8d56\u65e0 PU \u57f9\u517b\u57fa\u7b5b\u9009 \u2014\u2014 \u56e0 mtDNA \u7f3a\u9677\u7ec6\u80de\uff08\u03c1\u2070\u7ec6\u80de\uff09\u65e0\u6cd5\u81ea\u4e3b\u5408\u6210\u4e19\u916e\u9178\u548c\u5c3f\u82f7\uff0c\u4ec5\u878d\u5408\u7ec6\u80de\u53ef\u5b58\u6d3b\u3002\u4f46\u8be5\u65b9\u6cd5\u5b58\u5728\u7b5b\u9009\u5468\u671f\u957f\u3001\u7ec6\u80de\u6d3b\u6027\u4f4e\u7b49\u95ee\u9898\u3002<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u5fae\u6d41\u63a7\u6280\u672f\u7684\u4ecb\u5165\u5b9e\u73b0\u4e86\u4e09\u5927\u7a81\u7834\uff1a<strong>\u4e00\u662f<\/strong><strong> PU <\/strong><strong>\u8865\u5145\u6761\u4ef6\u4e0b\u7684\u9009\u62e9\u6027\u57f9\u517b<\/strong>\uff0c\u901a\u8fc7\u5fae\u901a\u9053\u7cbe\u51c6\u63a7\u5236\u7ec6\u80de\u5fae\u73af\u5883\uff0c\u65e0\u9700\u4f9d\u8d56\u8425\u517b\u7f3a\u9677\u7b5b\u9009\uff1b<strong>\u4e8c\u662f\u5355\u7ec6\u90e8\u5206\u878d\u5408<\/strong>\uff0c\u5229\u7528\u5fae\u6d41\u63a7\u82af\u7247\u7684\u7ec6\u80de\u914d\u5bf9\u7ed3\u6784\uff08CPS\uff09\u4e0e\u5fae\u96a7\u9053\u8bbe\u8ba1\uff0c\u5b9e\u73b0\u7ebf\u7c92\u4f53\u5b9a\u5411\u8f6c\u79fb\u800c\u907f\u514d\u7ec6\u80de\u6838\u878d\u5408\uff1b<strong>\u4e09\u662f\u9ad8\u6548\u7b5b\u9009\u4e0e\u9274\u5b9a<\/strong>\uff0c\u7ed3\u5408 PCR \u4e0e\u9650\u5236\u6027\u7247\u6bb5\u957f\u5ea6\u591a\u6001\u6027\uff08RFLP\uff09\u5206\u6790\uff0c\u5feb\u901f\u786e\u8ba4 mtDNA \u6765\u6e90\u3002<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u8fd9\u4e00\u6280\u672f\u4f9d\u8d56<strong><a href=\"https:\/\/www.dxfluidics.com\/en\/microfluidic-device\/pdms\u82af\u7247\u52a0\u5de5\u8bbe\u5907\/\">PDMS \u82af\u7247\u52a0\u5de5<\/a><\/strong>\u7684\u9ad8\u7cbe\u5ea6\u5fae\u7ed3\u6784 \u2014\u2014 \u901a\u8fc7\u8f6f\u5149\u523b\u6280\u672f\u5236\u5907\u7684\u5fae\u96a7\u9053\uff08\u957f\u5ea6 4-10\u03bcm\uff09\u4e0e\u7ec6\u80de\u53e3\u888b\uff0c\u4e3a\u4f9b\u4f53\uff08\u542b mtDNA \u7684 Pur\u1d63\u7ec6\u80de\uff09\u4e0e\u53d7\u4f53\uff08\u03c1\u2070\u7ec6\u80de\uff09\u63d0\u4f9b\u53ef\u63a7\u63a5\u89e6\u7a7a\u95f4\uff0c\u914d\u5408 HVJ-E \u75c5\u6bd2\u4ecb\u5bfc\u7684\u878d\u5408\u4f53\u7cfb\uff0c\u5b9e\u73b0\u9ad8\u6548\u7ebf\u7c92\u4f53\u8f6c\u79fb\u3002<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">2.&nbsp;&nbsp;&nbsp; \u6838\u5fc3\u5b9e\u9a8c\u6d41\u7a0b\uff1a\u4ece\u7ec6\u80de\u51c6\u5907\u5230\u6742\u5408\u7ec6\u80de\u6536\u83b7<\/h2>\n\n\n\n<h3 class=\"wp-block-heading\">1)&nbsp;&nbsp;&nbsp; \u4f9b\u4f53\u4e0e\u53d7\u4f53\u7ec6\u80de\u7684\u7cbe\u51c6\u5236\u5907<\/h3>\n\n\n\n<p \u7ec6\u80de\uff09\u3002\u03c1\u2070\u7ec6\u80de\u5236\u5907\u9700\u7ecf\u6eb4\u5316\u4e59\u952d\uff08EtBr\uff09\u5904\u7406 30-60 \u5929\uff0c\u6bcf\u65e5\u66f4\u6362\u57f9\u517b\u57fa\u4ee5\u6e05\u9664 mtDNA\uff0c\u540e\u7eed\u5728 PU \u57f9\u517b\u57fa\u4e2d\u57f9\u517b 1 \u4e2a\u6708\u4ee5\u4e0a\uff0c\u901a\u8fc7 PCR \u786e\u8ba4\u65e0 mtDNA \u6b8b\u7559\u3002<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u5173\u952e\u64cd\u4f5c\u5305\u62ec\uff1a<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>\u4f9b\u4f53\u7ec6\u80de\u9700\u8f6c\u5165\u8367\u5149\u6807\u8bb0\u57fa\u56e0\uff08\u5982 H2B-EGFP\uff09\uff0c\u53d7\u4f53\u7ec6\u80de\u8f6c\u5165 mCherry \u6807\u8bb0\uff0c\u4fbf\u4e8e\u540e\u7eed\u8ffd\u8e2a\uff1b<\/li>\n\n\n\n<li>\u786e\u4fdd\u4f9b\u4f53\u4e3a TK\u207a\u3001\u53d7\u4f53\u4e3a TK\u207b\uff0c\u901a\u8fc7 BrdU
